High-dose chemotherapy followed by autologous stem-cell transplantation versus non-myeloablative consolidation in primary CNS lymphoma (MATRix/IELSG43): a randomised phase 3 trial

This study aimed to compare the efficacy and safety of high-dose chemotherapy followed by autologous stem-cell transplantation (HCT-ASCT) versus non-myeloablative chemoimmunotherapy (R-DeVIC) as consolidation therapy in newly diagnosed primary CNS lymphoma (PCNSL) patients. The results demonstrated that HCT-ASCT significantly improved 3-year progression-free survival (78% vs. 51%) compared to R-DeVIC, establishing HCT-ASCT as the preferred consolidation strategy for eligible patients.
