Plozasiran for Severe Hypertriglyceridemia and Risk of Pancreatitis.

Listen to this summary

Researchers asked whether plozasiran, an siRNA therapy that silences APOC3 mRNA, can lower triglycerides and reduce acute pancreatitis risk in adults with severe hypertriglyceridemia. In two pooled, double-blind, placebo-controlled trials of 757 patients, plozasiran cut median triglycerides by about 79-81% from baseline at 12 months, versus placebo. Acute pancreatitis occurred at 1 versus 5 per 100 patient-years, with a rate ratio of 0.22 (P = 0.008), suggesting a meaningful reduction in pancreatitis risk.
